The Barossa will host a start and finish in the Santos Tour Down Under for the first time in 15 years on January 21.
The first stage will start at Nuriootpa and loop through the major towns three times before finishing at Angaston.
Cyclists will pass through Nuriootpa, Greenock, Seppeltsfield, Marananga, Tanunda, Bethany and Angaston, completing the Menglers Hill climb on the third lap of the 135km stage.
The Barossa Council expects to spend up to $70,000 on the event, including $35,000 to secure stage one and pay staff wages on the day.
